I have a main router Ax55 on 1st floors, a RE505x on 2nd floor as onemesh extender, could I use 1 more RE315 extender on 3rd floor to connect to RE505x on 2nd floor to make the mesh network?

You can but its recommended to connected both extenders to the main router for best performance.

As the workaround, you may connect the 2nd range extender to the 1st range extender via Ethernet cable and use the 2nd extender in the AP mode.
